Daya Hambat Pasta Gigi yang Mengandung Ekstrak Biji Kopi Robusta (Coffea canephora) terhadap Pertumbuhan Bakteri Porphyromonas gingivalis

Abstract


Porphyromonas gingivalis is one of the bacteria that causes periodontal disease. Toothpaste containing robusta coffee beans can be developed for the prevention and treatment of periodontal disease, because robusta coffee beans have antibacterial potential. The purpose of this study was to evaluate the antibacterial effect of robusta coffee bean extract toothpaste on the growth of Porphyromonas gingivalis bacteria. This research was an in vitro experimental research. Robusta coffee beans were extracted using the maceration method and formulated into toothpaste with a concentration of 6.25%; 12.5%; 25%; and 50%. Furthermore, an antibacterial test was carried out using the well-diffusion method on MHA media that had been inoculated with Porphyromonas gingivalis and the diameter of the inhibition zone formed around the wells was measured. The results showed that the zone of inhibition around the wells of the toothpaste was robusta coffee bean extract with a concentration of 12.5%; 25%; and 50%. The largest inhibition zone was indicated by toothpaste containing 50% concentration of robusta coffee bean extract, namely 19.18 + 0.18 mm. The higher the concentration of Robusta coffee bean extract, the greater the inhibition zone formed. It can be concluded that toothpaste containing robusta coffee bean extract is able to inhibit the growth of Porphyromonas gingivalis bacteria so that it can be developed as a toothpaste for the prevention and treatment of periodontal disease.

ABSTRAK

 

Porphyromonas gingivalis merupakan salah satu bakteri penyebab penyakit periodontal. Pasta gigi yang mengandung biji kopi robusta dapat dikembangkan untuk pencegahan dan perawatan penyakit periodontal, karena biji kopi robusta memiliki potensi antibakteri. Tujuan dari penelitian ini adalah untuk mengevaluasi efek antibakteri pasta gigi esktrak biji kopi robusta terhadap pertumbuhan bakteri Porphyromonas gingivalis. Penelitian ini adalah penelitian eksperimental in vitro. Biji kopi robusta diekstrak menggunakan metode maserasi dan diformulasikan menjadi pasta gigi dengan konsentrasi 6,25%; 12,5%; 25%; dan 50%. Selanjutnya dilakukan uji antibakteri menggunakan metode difusi sumuran pada media MHA yang telah diinokulasikan Porphyromonas gingivalis dan dilakukan pengukuran diameter zona hambat yang terbentuk di sekitar lubang sumuran. Hasil penelitian menunjukkan bawa zona hambat di sekitar lubang sumuran pasta gigi ekstrak biji kopi robusta konsentrasi 12,5%; 25%; dan 50%. Zona hambat terbesar ditunjukkan oleh pasta gigi yang mengandung ekstrak biji kopi robusta konsentrasi 50% yaitu 19,18 + 0,18 mm. Semakin tinggi konsentrasi ekstrak biji kopi robusta, semakin besar juga zona hambat yang terbentuk. Dapat disimpulkan bahwa pasta gigi yang mengandung ekstrak biji kopi robusta mampu menghambat pertumbuhan bakteri Porphyromonas gingivalis sehingga dapat dikembangkan sebagai pasta gigi untuk pencegahan dan perawatan penyakit periodontal.
