Isolasi Fungi Endofit dari Buah dan Daun Belimbing Wuluh (Averrhoa bilimbi L.)

Abstract

Isolation in this study is the first step to determine the presence of endophytic fungi in the fruit and leaves of starfruit (Averrhoa bilimbi L.). The aim of this study was to determine whether there were endophytic fungi in the fruit and leaves of starfruit. The type of research used was descriptive with laboratory tests to isolate endophytic fungi from the fruit and leaves of starfruit. The sample used were fruit and leaves of starfruit. This research was carried out for 3 months, from January 9 to April 9, 2021 at the Microbiology Laboratory, FMIPA, Cendrawasih University, Jayapura. Data collection was carried out in the following stages: (1) isolation of endophytic fungi using the surface sterilization method and (2) conventional identification of endophytic fungi by observing macroscopic and microscopic morphological characters. Based on the isolation results, two isolates were obtained from fruit and three isolates from leaves. The conclusion of this study is that the endophytic fungi that have been isolated are the Genus Penicillium sp, Rhizopus sp, Aspergillus sp, Phoma sp and one isolate that has not been identified.

ABSTRAK

Isolasi dalam penelitian ini merupakan langkah awal untuk mengetahui keberadaan fungi endofit dalam buah dan daun belimbing wuluh (Averrhoa bilimbi L.). Tujuan dalam penelitian ini adalah untuk mengetahui ada tidaknya fungi endofit pada buah dan daun belimbing wuluh. Jenis penelitian yang digunakan adalah deskriptif dengan uji laboratorium untuk mengisolasi jamur endofit dari buah dan daun belimbing wuluh. Sampel yang digunakan adalah buah dan daun belimbing wuluh. Penelitian ini dilaksanakan selama 3 bulan, dari tanggal 09 Januari sampai dengan 09 April 2021 di Laboratorium Mikrobiologi FMIPA Universitas Cendrawasih Jayapura. Pengumpulan data dilakukan dengan tahap sebagai berikut: (1) isolasi fungi endofit dengan menggunakan metode sterilisasi permukaan dan (2) identifikasi fungi endofit secara konvesional yaitu dengan mengamati karakter morfologi secara makroskopis dan mikroskopis. Berdasarkan hasil isolasi diperoleh dua isolat yang berasal dari buah dan tiga isolat dari daun. Kesimpulan penelitian ini adalah fungi endofit yang berhasil diisolasi adalah Genus Penicillium sp, Rhizopus sp, Aspergillus sp, Phoma sp dan satu isolat yang belum teridentifikasi.
