Potensi Fraksi Etil Asetat Daun Nipah (Nypa fruticans. Wurmb) Terhadap Aktivitas Antihiperglikemi, Sekresi Insulin dan Regenerasi Sel β-Pankreas Pada Tikus Yang Diinduksi STZ
Abstract
Diabetes mellitus (DM) is a metabolic syndrome in the endocrine system characterized by hyperglycemia caused by reduced insulin secretion, insulin resistance or both. Nypa fruticans is one of the plants used in the traditional treatment of diabetes. Flavonoid content such as quercetin, eriodictyol, catechin, hydroquinone, apigenin, camphorol, anthocyanin and rutin support the potential of nipah plants as antidiabetics. This study was designed to evaluate the antihyperglycemic potential of the ethyl acetate fraction by observing the increase in insulin secretion, improvement of β-pancreatic cells and increase in the number of β-pancreatic cells. Ethyl acetate fraction of nipah leaves with doses of 125, 250 and 500 mg/kgBW were given to diabetic rats induced by streptozotocin (STZ) 65 mg/kgBW and glibenkalmide 045 mg/kgBW as a comparison for 21 days with parameters observed including serum insulin levels, and pancreatic histopathology. Nipah leaf ethyl acetate fraction with doses of 250 and 500 mg/kgBW caused a significant decrease (p < 0.05) in blood glucose levels by significantly increasing (p < 0.05) serum insulin levels when compared to negative controls. In histopathological studies of the pancreas, the ethyl acetate fraction showed a regeneration effect on pancreatic β-cells by reducing the degree of insulitis and significantly increasing (p < 0.05) the number of pancreatic β-cells of the islets of Langerhans when compared to negative controls. Based on these results, the ethyl acetate fraction of nipah leaves showed antihyperglycemic activity by regenerating pancreatic β-cells and increasing the number of pancreatic β-cells, thereby increasing serum insulin levels.
