Pengaruh Ekstrak Etanol Biji Alpukat (Persea americana Mill) terhadap Kadar Glukosa Darah dan Ekspresi SOD2 Tikus Diabetes Militus

Abstract


 Diabetes mellitus is a chronic disease caused by damage to the pancreas. This research was conducted with the aim of looking at the potential of avocado seed extract (Persia americana Mill) as an alternative medicine for diabetes. This research was an experimental study using mice as experimental animals. In the in vivo test, mice were divided into 5 groups, namely: group 1 (healthy or untreated group) and groups 2, 3, 4 and 5 which were injected with streptozotocin (STZ) at a dose of 30 mg/kbBB intraperitoneally. Group 2 was a group of sick mice without therapy, while groups 3, 4, and 5 were groups of sick mice treated with ethanol extract of avocado seeds, at doses of 300, 350 and 400 mg/kgBW respectively for 14 days. In each group, blood glucose levels and SOD2 expression were examined using immunohistochemical techniques. Next, an analysis of differences in blood glucose levels and SOD2 expression between groups was carried out using the ANOVA test. The results of the analysis showed that therapy with ethanol extract of avocado seeds (P. americana Mill) showed a less significant reduction in blood glucose levels, but there was a significant increase in SOD2 expression (p <0.05). It was concluded that the ethanol extract of avocado seeds has potential as a basic ingredient for antidiabetic drugs.

ABSTRAK

 

Diabetes melitus merupakan penyakit kronis yang disebabkan oleh kerusakan pankreas. Penelitian ini dilakukan dengan tujuan untuk melihat potensi ekstrak biji alpukat (Persia americana Mill) sebagai obat alternatif diabetes. Penelitian ini merupakan studi eksperimental menggunakan tikus sebagai hewan coba. Pada uji in vivo, tikus dibagi dalam 5 kelompok yaitu: kelompok 1 (kelompok sehat atau tanpa perlakuan) dan kelompok 2, 3, 4 dan 5 yang diinjeksi dengan streptozotocin (STZ) dosis 30 mg/kbBB di intraperitoneal. Kelompok 2 adalah kelompok tikus sakit tanpa terapi, sedangkan kelompok 3, 4, dan 5 merupakan kelompok tikus sakit yang diterapi dengan ekstrak etanol biji alpukat, di mana dosis masing-masing 300, 350 dan 400 mg/kgBB selama 14 hari. Pada masing-masing kelompok dilakukan pemeriksaan kadar glukosa darah dan ekspresi SOD2 dengan teknik imunohistokimia. Selanjutnya dilakukan analisis perbedaan kadar glukosa darah dan ekspresi SOD2 antar kelompok menggunakan uji ANOVA. Hasil analisis menunjukkan bahwa terapi dengan ekstrak etanol biji alpukat (P. americana Mill) menunjukkan adanya penurunan kurang signifikan pada kadar glukosa darah, tetapi adanya peningkatan ekspresi SOD2 secara signifikan (p <0,05). Disimpulkan bahwa ekstrak etanol biji alpukat berpotensi sebagai bahan dasar obat antidiabetes.
