Efek Kombinasi Ekstrak Pegagan (Centella Asiatica) dan Ikan Gabus (Channa Striata) terhadap Fungsi Kognitif dan Tingkat Stres Oksidatif pada Tikus Model Penuaan

Abstract


Increased oxidative stress can accelerate the aging process. This process is also accompanied by a decrease in the body's ability to overcome the effects of free radicals. Spatial learning and memory behavior with the Morris Water Maze test were carried out to determine whether the mice had cognitive impairment. MDA is a product of lipid peroxidation which is used as a marker of oxidative stress. This study aimed to determine the effect of a combination of gotu kola and snakehead fish extracts in reducing oxidative stress, cognitive function and brain histopathology in aging model mice. The design of this research was a post test only control group design. There were 7 sample groups, each consisting of 4 Wistar rats (n = 28). The control group (K0) was only given standard feed, the negative control group was induced with D-galactose 150 mg / kg / body weight, group P1 = Gotu Kola (Centella Asiatica) extract group 300 mg/kgBW/day and injected with D-galactose 150 mg/kgBW /day, P2 = Group of snakehead fish extract (Channa Striata) 300 mg/kg/day and injected with D-galactose 150 mg/kgBW/day, P3 = Group of gotu kola (Centella Asiatica) extract and snakehead fish extract (Channa Striata) 100/day 300 mg/kg/day and injected D-galactose 150 mg/kgBW/day, P4 = Gotu kola (Centella Asiatica) extract group and snakehead fish extract (Channa Striata) 300/300 mg/kg and injected D-galactose 150 mg/kgBW /day, P5 = Pegagan (Centella Asiatica) and snakehead fish (Channa Striata) extract groups 600/300 mg/kg and injected with D-galactose 150 mg/kgBW/day, treatment was given for 6 weeks. The collected data was analyzed using the One Way Annova test. The analytical results showed p-value <0.05 for cognitive function, p <0.05 for oxidative stress (MDA) levels, and p <0.05 for the histopathological picture of the rat brain. It can be concluded that in aging model mice, administration of a combination of gotu kola and snakehead fish extracts can reduce stress levels, improve cognitive function and increase neurogenesis.

Peningkatan stress oksidatif dapat mempercepat proses penuaan. Proses ini juga disertai penurunan kemampuan tubuh dalam mengatasi efek radikal bebas. Pembelajaran spasial dan perilaku memori dengan tes morris water maze dilakukan untuk menentukan apakah tikus mengalami gangguan kognitif. MDA adalah produk dari peroksidasi lipid yang digunakan sebagai marker stress oksidatif. Penelitian ini bertujuan untuk mengetahui efek dari kombinasi ekstrak pegagan dan ikan gabus dalam mengurangi stress oksidatif, fungsi kognitif dan gambaran histopatologi otak  pada tikus model penuaan. design penelitian ini adalah post test only control group design. Ada 7 kelompok sampel yang masing-masing terdiri atas 4 tikus galur wistar (n = 28). Kelompok kontrol (K0) hanya diberikan pakan standar, kelompok kontrol negatif diinduksi dengan D-galaktosa 150 mg / kg / bb, kelompok P1 = Kelompok ekstrak pegagan (Centella Asiatica) 300 mg/kgBB/hari dan diinjeksi D-galaktosa 150 mg/kgBB/hari, P2 = Kelompok ekstrak ikan gabus (Channa Striata) 300 mg/kg/hari dan diinjeksi D-galaktosa 150 mg/kgBB/hari, P3 = Kelompok ektrak pegagan (Centella Asiatica) dan ektrak ikan gabus (Channa Striata) 100/300 mg/kg/hari dan diinjeksi D-galaktosa 150 mg/kgBB/hari, P4 = Kelompok ekstrak pegagan (Centella Asiatica) dan ekstrak ikan gabus (Channa Striata) 300/300 mg/kg dan diinjeksi D- galaktosa 150 mg/kgBB/hari, P5 = Kelompok ekstrak pegagan (Centella Asiatica) dan ikan gabus (Channa Striata) 600/300 mg/kg dan diinjeksi D-galaktosa 150 mg/kgBB/hari, perlakuan diberikan selama 6 minggu. Data yang telah terkumpul dianalis menggunakan uji One Way Annova. Hasil analis menunjukkan nilai p <0,05 untuk fungsi kognitif, p <0.05 untuk tingkat stress oksidatif (MDA), dan p <0.05 untuk gambaran histopatologi otak tikus. Dapat disimpulkan bahwa pada tikus model penuaan, pemberian kombinasi ekstrak pegagan dan ikan gabus dapat menurunkan tingkat stress, dan dapat meningkatkan fungsi kognitf serta dapat meningkatkan neurogenesis.
